Overview

Semax is a synthetic heptapeptide (Met-Glu-His-Phe-Pro-Gly-Pro) derived from ACTH(4-10), developed in Russia in the 1980s. REGULATORY: Approved in Russia and on Russian List of Vital & Essential Drugs (Dec 2011). NOT FDA-approved in US or most Western countries. BEST EVIDENCE: Stroke recovery - multiple RCTs (n=110+ patients total) showing increased BDNF, improved motor function, and faster recovery. COGNITIVE ENHANCEMENT: Limited evidence - one study showed 30% improved attention in healthy volunteers, but most studies are in Russian with limited Western validation. Mechanism: Enhances BDNF (20-24hr duration from Pro-Gly-Pro modification), activates neurotrophin transcription. PRACTICAL: Convenient intranasal administration (vs IV Cerebrolysin). SAFETY: Excellent profile from decades of Russian use - no addiction, withdrawal, or major side effects. Better evidence than BPC-157/TB-500/Dihexa but less rigorous than FDA-approved therapies.
