Overview
Selank (TP-7) is a synthetic heptapeptide derived from the naturally occurring immunomodulatory peptide tuftsin (Thr-Lys-Pro-Arg) with the addition of a Pro-Gly-Pro sequence for enhanced stability and CNS activity. Developed at the Institute of Molecular Genetics of the Russian Academy of Sciences, Selank was approved in Russia in 2009 as an intranasal anxiolytic and nootropic medication. Its mechanism involves modulation of GABAergic neurotransmission, serotonin metabolism, and brain-derived neurotrophic factor (BDNF) expression. Russian clinical studies demonstrated anxiolytic effects comparable to benzodiazepines but without sedation, cognitive impairment, or dependence — making it an attractive alternative for anxiety disorders. Selank also demonstrates nootropic properties, improving attention, memory, and learning in clinical trials. Additionally, as a tuftsin analog, Selank retains immunomodulatory properties, enhancing IL-6 and affecting immunoglobulin gene expression. Selank is administered intranasally as a 0.15% solution, providing rapid CNS access through the nasal mucosa. The half-life is very short (approximately minutes), but clinical effects persist for hours due to downstream signaling changes. Selank has a clean side effect profile in clinical use, with no reported cases of dependence, withdrawal, or significant adverse events.
