Overview
Resveratrol (3,5,4'-trihydroxystilbene) is a naturally occurring polyphenol found in grape skins, red wine, and Japanese knotweed (Polygonum cuspidatum). Its dermatological significance stems from multiple well-characterized mechanisms: activation of SIRT1 (a NAD+-dependent deacetylase central to longevity pathways), potent NF-kB inhibition, and antagonism of the aryl hydrocarbon receptor (AhR) which mediates pollution-induced skin aging. Multiple RCTs have demonstrated topical resveratrol (0.4-1%) reduces photoaging markers, decreases erythema, and improves skin texture. Oral supplementation (75-500 mg/day) has shown promise for improving skin elasticity and dermal collagen density in clinical trials. However, resveratrol has poor aqueous solubility and rapid metabolic clearance, requiring advanced delivery systems (nanoencapsulation, liposomal) for optimal topical efficacy. Its combination with vitamin E enhances photostability and antioxidant synergy.
