Overview
MK-677 (Ibutamoren) is an oral, non-peptide ghrelin mimetic (growth hormone secretagogue) that stimulates GH release from the pituitary gland without affecting cortisol levels. A 12-month randomized clinical trial in healthy older adults (n=65) demonstrated that daily MK-677 increased GH secretion to youthful levels and fat-free mass (lean body mass) by approximately 1.5 kg compared to placebo. However, it did NOT significantly improve muscle strength, physical function, or quality of life measures. Other controlled studies in healthy volunteers confirmed oral MK-677 increases GH (97%) and IGF-1 (40-50%) levels. Small studies show beneficial effects on sleep architecture, including 50% increase in Stage IV (deep) sleep and 20% increase in REM sleep. Phase 2 trials evaluated MK-677 for conditions like fibromyalgia. CRITICAL SAFETY NOTICE: MK-677 is NOT approved by the FDA or other regulatory agencies for any therapeutic use. Long-term safety has not been established. Potential risks include changes in blood sugar, insulin sensitivity, and metabolic effects. Prohibited by WADA and sports authorities.
