GHK-Cu (Copper Peptide) for Skin, Hair & Anti-Aging — Skin & Hair
Topical cosmetic peptide with limited published evidence - NOT FDA-approved for anti-aging; most cited studies unpublished.
Overview
GHK-Cu (glycyl-L-histidyl-L-lysine-copper) is a naturally occurring tripeptide available as OTC cosmetic ingredient. CRITICAL EVIDENCE STATUS: ❌ NO FDA approval for cosmetic/anti-aging use. PUBLISHED STUDIES: Only ONE peer-reviewed positive study exists (Abdulghani 1998) - applied to THIGH SKIN (not face), 1 month duration. The most frequently cited facial studies (Leyden 2002) are UNPUBLISHED conference abstracts, never peer-reviewed after 23+ years. One rigorous study (CO2 laser) showed NO objective improvement in wrinkles. Recent 'studies' are company press releases, not peer-reviewed research. Mechanism is promising (stimulates collagen in vitro), safety is excellent (decades of use), but clinical evidence is cosmeceutical-grade: strong marketing, weak published data. BETTER ALTERNATIVES: FDA-approved tretinoin has 40+ years of large RCT data published in top-tier journals.
Indications
- ⚠️ Limited evidence for facial anti-aging (most cited studies unpublished)
- Skin aging, fine lines, and wrinkles (cosmeceutical-grade evidence)
- Wound healing post-procedure (clinical support for adjunct use)
- Hair thinning (anecdotal, limited clinical data)
- NOT first-line for photoaging - consider tretinoin (FDA-approved)
Mechanism of Action
Natural plasma levels decline from ~200 ng/mL (age 20) to ~80 ng/mL (age 60)
Dosing
| Compound | Dose | Frequency | Notes |
|---|---|---|---|
| GHK-Cu 0.05-0.5% | Apply to face/neck | Once or twice daily | OTC cosmetic. Limited published evidence for facial anti-aging. |
| GHK-Cu 0.5-2% | Apply to targeted areas | Once or twice daily | Higher concentration. No dose-response studies in peer-reviewed literature. |
| GHK-Cu | 1-2 mg | Once daily | ⚠️ ZERO published RCTs on injectable GHK-Cu in humans. All data extrapolated. |
| GHK-Cu 2mg in 2mL saline | Apply post-needling | Every 2 weeks | Professional treatment. Wound healing evidence supports adjunct use. |

Safety & Contraindications
- ✅ Excellent safety profile (decades of cosmetic use, well-tolerated)
- ✅ Naturally occurring peptide in human plasma
- ⚠️ NOT FDA-approved for anti-aging (OTC cosmetic ingredient only)
- Contraindicated in Wilson's disease (copper metabolism disorder)
- Must supplement with zinc (15-30mg daily) to prevent copper/zinc imbalance
- 'Copper uglies': Excess use may paradoxically increase MMP-1 and cause collagen breakdown
- ⚠️ INJECTABLE: ZERO published RCTs in humans - all injectable data from animal studies
