Overview
Bakuchiol is a plant-derived meroterpene that has emerged as a functional retinol analog with comparable efficacy for photoaging and acne but significantly fewer adverse effects. A landmark double-blind RCT published in the British Journal of Dermatology (2019, PMID: 30516284) demonstrated that 0.5% bakuchiol applied twice daily produced statistically comparable improvements in wrinkle surface area, hyperpigmentation, and overall photodamage versus 0.5% retinol over 12 weeks, with significantly less scaling and stinging. Unlike retinoids, bakuchiol does not bind RAR/RXR receptors but instead modulates gene expression through alternative pathways including upregulation of type I, III, and IV collagen genes. It exhibits additional anti-inflammatory activity via inhibition of NF-kB and IL-6 pathways. Bakuchiol is photostable and does not increase UV sensitivity, making it suitable for daytime use. Pregnancy safety data remain limited, but in vitro genotoxicity and reproductive toxicity screens are negative.
