Overview
Apolipoprotein E (APOE) has three common alleles: ε2, ε3, and ε4. APOE4/4 homozygotes (2% of population) have 8-12x higher lifetime Alzheimer's risk; APOE4/3 heterozygotes (24% of population) have 2-4x increased risk. APOE4 also accelerates cardiovascular disease, worsens outcomes with head trauma, and affects response to certain dietary interventions (e.g., saturated fat raises LDL more in APOE4 carriers). APOE2/2 is protective against Alzheimer's.
