5-MeO-DMT (Bufo Alvarius / Synthetic) — Psychedelics & Neuroplasticity
The most potent naturally occurring psychedelic by weight — produces profound ego dissolution, non-dual consciousness, and lasting psychological benefit from a single 15-minute experience.
Overview
5-methoxy-N,N-dimethyltryptamine (5-MeO-DMT) is found in the secretion of the Sonoran Desert toad (Bufo alvarius / Incilius alvarius) and in several plants. It is approximately 4-6x more potent than DMT and produces overwhelming non-dual consciousness experiences lasting 15-30 minutes when vaporized. Johns Hopkins researchers published the first prospective study (2019, Psychopharmacology, n=362) showing that a single 5-MeO-DMT session produced significant reductions in depression (66% reduced symptoms) and anxiety (57% reduced symptoms) with 80% rating it as one of the most meaningful experiences of their lives. The mechanism is distinct from classical psychedelics — 5-MeO-DMT acts primarily at 5-HT1A receptors (not 5-HT2A) and at sigma-1 receptors. Synthetic pharmaceutical-grade 5-MeO-DMT (produced without harming toads) is preferred ethically over toad secretion (Bufo alvarius populations are threatened by over-collection). Beckley Psytech is conducting Phase 1/2 trials with synthetic 5-MeO-DMT (BPL-003) for treatment-resistant depression.
Indications
- Treatment-resistant depression (preliminary human evidence)
- Existential anxiety and end-of-life distress
- Addiction recovery support
- PTSD (anecdotal and preliminary data)
Mechanism of Action
Unlike classical psychedelics, 5-MeO-DMT acts primarily at 5-HT1A (serotonin autoreceptors) rather than 5-HT2A, producing sedation, anxiolysis, and anti-depressant-like neural patterns distinct from classical psychedelics
Dosing
| Compound | Dose | Frequency | Notes |
|---|---|---|---|
| 5-MeO-DMT (synthetic) | 5-15 mg vaporized | Single session; months to years between experiences | Onset within 30-60 seconds; peak 5-15 minutes; duration 15-30 minutes total; full supervision required |
Evidence Grade
GRADE C
Safety & Contraindications
- Overwhelming experience — potential for psychological trauma without adequate preparation and support
- Vomiting can occur — position to prevent aspiration
- Serotonin syndrome risk if combined with MAOIs or serotonergic medications
- Seizure risk reported, especially in predisposed individuals
- Cardiovascular effects during session — avoid in cardiac disease
- Legal as toad secretion in some jurisdictions; synthetic 5-MeO-DMT is Schedule I in USA
- Strong advocate for synthetic over toad-sourced due to toad conservation concerns
